Principal Scientist, Structural Biology & Protein Design

Job Scope & Purpose:

Our exclusive client is seeking an experienced individual to lead structure biology efforts for our newly discovered protein systems group. Structural biology is a key component for translating our discovery efforts into therapeutic applications. As a Principal Scientist, you will be responsible for developing structural biology pipelines through both internal initiatives and CROs. Additionally, you are also expected to use structural knowledge to provide mechanistic insights of protein systems, and work with protein engineers to guide structure-based designs and evaluate protein engineering and product development projects. Previous industry experiences and management of a team and CROs are highly preferred.

Primary Job Responsibilities:

  • Develop the current structural biology program to achieve high-resolution structures for multiple CRISPR systems
  • Leverage the structure information to support protein engineering and product development efforts
  • Provide structure-function insights to guide biochemical and mechanistic characterizations
  • Provide leadership and expertise in structure-based design and modeling
  • Perform protein expression and purification, crystallizing, X-ray structure determination and/or Cryo-EM structure determination through both internal initiatives and CROs

Primary Job Requirements:
  • A Ph.D in structure biology or related field with additional 2+ years of experience in industry
  • Demonstrated ability to solve multiple protein structures and tackle difficult problems with Cryo-EM and X-ray crystallography.
  • Extensive knowledge of protein purification, biophysical characterization, protein structure and function, with familiarity in structural modeling and design
  • Excellent project management and organizational skills
  • Basic knowledge of CRISPR gene editing technologies, DNA nuclease structure, function and engineering
  • Proficiency with biochemical (enzymatic activity, protein-nucleic acid interactions) and bioanalytical methods (such as DLS, DSF, SEC-MALS) to evaluate protein quality/stability and determine optimal protein/RNP formulations.
  • Direct experience in protein engineering and methods for directed evolution in mammalian cell systems is strongly preferred
  • Familiarity with standard mammalian cell culture techniques, including methods for gene delivery including and lenti-viral pooled screening approaches is a plus
  • Familiarity with CMC manufacturing best practices is a plus but we are willing to train an otherwise qualified candidate
