The lab of Haruki Hasegawa in Biologics Department at Amgen South San Francisco is seeking a postdoctoral fellow to investigate various intracellular protein phase separation events and cellular phenotypes induced by the overexpression of condensation-prone secretory cargo proteins. The labs research interests include the elucidation of cargo-specific biosynthetic requirements for various classes of therapeutically important proteins (PMIDs: , , , , ), characterization of intracellular protein crystallization and protein phase separation in secretory pathway organelles (PMIDs: , , , , , ), understanding the relationships between overexpression-induced cell phenotypes and antibodys solubility characteristics (PMIDs: , , , , ), and biotechnological application of intra-ER protein inclusion bodies formation.

The initial research projects of the selected postdoctoral fellow are: (1) characterizing the roles of post-translational modifications and protein-protein interactions in intra-ER protein crystallization event; (2) elucidating the solubility and condensation propensity of a human IgG that induces LLPS protein droplet in the ER lumen during overexpression; (3) exploring the merits of identified 5-UTRs and various effector genes in enhancing difficult-to-express secretory protein production, to list a few potential projects.

The postdoctoral fellow is expected to develop an independent line of research based on initial projects and to present at scientific conferences and publish in leading journals. The successful candidate will operate in a highly collaborative environment, with opportunities to interact with other postdoctoral fellows, scientists, and drug discovery teams while having access to state-of-the art protein expression technologies and resources at core facilities.

The postdoctoral position is for 3 years with the option to extend for an additional year.

Basic qualifications

Ph.D. at the time of appointment

Preferred qualifications

Ph.D. in molecular and cell biology, biochemistry, or related life science field

Solid publication record and strong oral and written communication skills

Extensive cell imaging experience in immunofluorescent microscopy (EPI, confocal, high content, etc)

Proficient skills in flow cytometry, cell sorting, intracellular signaling assays, protein purification, and chromatography

Cell phenotype morphometric analysis using algorithms and machine learning

Solid knowledge on secretory protein biosynthesis, folding, assembly, trafficking, and secretion

Familiarity with various stress response signaling pathways and associated cell-based assay methods

Research experience in the following areas:

- Intracellular protein phase separation, protein crystallization, or protein condensation

- Cell-based assay development and signaling pathway elucidation

- Recombinant antibody production and expression optimization

- Biochemical and biophysical characterization of purified recombinant antibodies
